Abstract
The goal of this research was to determine the Alu TPA-25 DNA fingerprint of my family members in order to construct a genetic pedigree. A non-invasive techniq ue was used to acquire buccal cells from 9 experimental subjects. Genomic DNA was released from the cells, mixed with specific primers, and subjected to the polymerase chain reaction (PCR) in order to amplify the Alu TP A-25 locus of chromosome 8. The PCR products were then electrophoresed on a 1.5% agarose gel to reveal the subject's genotype with respect to the Alu TPA-25 element.
Optimal amounts of PCR product were obtained by using the Amersham-Pharmacia Biotech Ready-to-Go Bead® system. Results with this system showed that I am homozygous recessive for the Alu TP A-25 allele. Similarly, all of my family members were also homozygous recessive. I used this information to create a pedigree chart of the Alu TPA-25 genotypes for my family.
