Abstract
The overall goal of the research was to identify DNA-binding proteins that might be involved in transcriptional regulation of phaZ. Specific goals were the following: 1) to construct DNA probes for use in EMSA; 2) to produce cell extracts of Streptomyces sp. 5A grown under inducing and repressing conditions; 3) to perform the EMSA technique to identify DNA-protein complexes. All goals were met and the EMSA technique was conducted three times. In the third EMSA procedure a possible shift was noted in PHB cell extracts collected after 29 hours of incubation.
