Abstract
This project involved using a lambda FIX II bacteriophage clone already constructed from a previous experiment to infect host Escherichia coli cells through lysis. The goal of the project was to isolate the lamda DNA contained inside the capsid of the bacteriophage. The bacteriophage's DNA was of interest because it contained an insert of horse DNA. Methods used to isolate the phage DNA included phage dilutions, plate lysates, liquid lysate cultures, ammonium sulfate precipitation, QIAGEN column purification, ethanol precipitation, and agarose gel electrophoresis.
